Guildrun hands out new heroes, items and relics as you play rather than all at once. Nine mastery bundles unlock at set XP thresholds, each adding a hero and the items and relics that fit the way that hero wants to play.
This page lists every bundle, the XP it takes to reach it and everything it adds, plus the XP each win and each completed stage is worth.
Heroes arrive in mastery bundles unlocked by earning XP, which you gain from winning encounters, completing stages and finishing runs.
